Overview

Axitinib + nivolumab is a combination regimen of two agents: axitinib, an oral small molecule tyrosine kinase inhibitor (TKI) that selectively inhibits vascular endothelial growth factor receptors (VEGFR) 1, 2, and 3, and nivolumab, a monoclonal antibody that targets the programmed death 1 (PD-1) immune checkpoint on T cells. Used together, they synergistically inhibit angiogenesis (tumor blood vessel growth) and enhance T-cell mediated anti-tumor immune response. This combination is under investigation and clinical use, particularly in metastatic renal cell carcinoma (mRCC) and in other advanced solid tumors, especially after progression on other therapies. The mechanistic rationale includes direct tumor growth suppression by VEGFR inhibition and activation of cytotoxic immune responses by PD-1 blockade.
