Axitinib + paclitaxel + carboplatin is a multi-agent chemotherapy regimen combining three drugs with distinct mechanisms of action, used primarily in oncology clinical trials. Axitinib is an oral small molecule tyrosine kinase inhibitor that selectively inhibits vascular endothelial growth factor receptors (VEGFR) 1, 2, and 3, thereby blocking angiogenesis required for tumor growth. Paclitaxel is a microtubule-stabilizing agent that disrupts mitosis by promoting tubulin polymerization and inhibiting depolymerization, leading to cell cycle arrest and apoptosis in rapidly dividing cells. Carboplatin is a platinum-based chemotherapeutic that forms DNA crosslinks, inhibiting DNA replication and transcription which induces apoptosis in cancer cells. This combination has been investigated particularly for BRAF wild-type metastatic melanoma but may be considered for other solid tumors as well[1][2][3]. The regimen has demonstrated tolerability with expected hematologic toxicities typical of cytotoxic chemotherapy[1][2].
