Overview

Axitinib + TRC105 is an investigational combination therapy for metastatic renal cell carcinoma (mRCC). Axitinib is a small molecule tyrosine kinase inhibitor that targets vascular endothelial growth factor receptors (VEGFR) and inhibits angiogenesis. TRC105 (carotuximab) is a chimeric IgG1 monoclonal antibody against endoglin (CD105), a receptor highly expressed on proliferating endothelial cells and certain cancer stem cells, implicated in angiogenesis and resistance to VEGF inhibitors. The strategy of combining these two drugs aims to overcome resistance to VEGF-targeted therapies by dual inhibition of VEGF and endoglin pathways. While phase 1 studies showed the combination is tolerable and demonstrated preliminary antitumor activity in previously treated mRCC patients, a phase 2 study did not show benefit over axitinib monotherapy in advanced clear cell renal cell carcinoma[1][2][4][5].
