AXP107-11 + gemcitabine is a combination therapy for pancreatic cancer, consisting of the genistein analogue AXP107-11 and the nucleoside analog chemotherapeutic gemcitabine. AXP107-11 is a multi-component crystalline form of genistein, designed for improved solubility and oral bioavailability compared to natural genistein. The combination was developed to overcome chemoresistance and enhance the efficacy of gemcitabine in pancreatic ductal adenocarcinoma. Preclinical and clinical studies show that AXP107-11 sensitizes tumor cells to gemcitabine via activation of the G-protein-coupled estrogen receptor (GPER1), leading to synergistic antitumor effects. Phase I and II trials demonstrated favorable safety, pharmacokinetic profile, and evidence of synergistic chemoenhancement in subsets of patients with pancreatic cancer[1][2][3][4][5][6][7].
