Azacitidine + cisplatin is a combination of two chemotherapeutic agents used experimentally in oncology. Azacitidine is a pyrimidine nucleoside analogue that acts as a DNA methyltransferase inhibitor, leading to hypomethylation of DNA and reactivation of silenced genes involved in tumor suppression. It also incorporates into RNA and DNA, disrupting their function and inhibiting protein synthesis[1][5]. Cisplatin is a platinum-based chemotherapy agent that forms DNA crosslinks, resulting in apoptosis by interfering with DNA replication and transcription[2]. The combination has been studied for its potential synergistic antitumor effects, particularly in overcoming chemoresistance in cancers such as hormone-refractory prostate cancer and germ cell tumors. Preclinical studies show that azacitidine can sensitize tumor cells to the cytotoxic effects of cisplatin by modulating gene expression related to cell cycle arrest and apoptosis[4][7].
