Overview

CACAG-VEN is an investigational multi-drug combination regimen developed by the Chinese PLA General Hospital for the treatment of acute myeloid leukemia (AML). The regimen integrates five active pharmaceutical ingredients: the BCL-2 inhibitor venetoclax, the hypomethylating agent azacitidine, the chemotherapy agents cytarabine and aclacinomycin (aclarubicin), and the histone deacetylase (HDAC) inhibitor chidamide, typically supplemented with granulocyte colony-stimulating factor (G-CSF). By targeting multiple oncogenic pathways—including DNA methylation, histone acetylation, apoptosis regulation, and DNA synthesis—the regimen aims to overcome resistance and improve remission rates in various AML populations. It is currently being evaluated in Phase 3 clinical trials as a first-line treatment for fit elderly patients with newly diagnosed AML, comparing its efficacy and safety against the standard "3+7" chemotherapy regimen.
