AZD0156 + olaparib is an experimental combination therapy consisting of AZD0156, a potent and selective oral inhibitor of ataxia-telangiectasia mutated (ATM) kinase, and olaparib, an approved oral poly(ADP-ribose) polymerase (PARP) inhibitor. The rationale for combining AZD0156 with olaparib is based on synergistic anti-tumor activity through dual inhibition of DNA damage repair pathways: AZD0156 impedes ATM signaling and DNA repair, thereby potentiating the cytotoxic effect of olaparib-induced DNA damage by further blocking repair of double-strand breaks. Preclinical evidence demonstrates enhanced efficacy in various cancer cell lines and xenograft models, particularly in ATM-deficient and BRCA-mutated cancers. This combination is currently being evaluated in early-phase clinical trials for advanced solid tumors to assess safety, tolerability, pharmacokinetics, and preliminary efficacy[1][2][3][5][6].
