Overview

**AZD1656 + metformin** is an investigational combination therapy comprised of AZD1656, a small molecule glucokinase activator, and metformin, a first-line oral antidiabetic drug. AZD1656 acts by activating glucokinase, an enzyme critical for glucose sensing in pancreatic beta cells and glucose utilization in the liver, thereby enhancing insulin-mediated glucose uptake and lowering blood glucose levels. Metformin reduces hepatic glucose production and improves insulin sensitivity, primarily lowering fasting plasma glucose. The combination has been studied for its synergistic potential to improve glycemic control in patients with type 2 diabetes mellitus not adequately controlled with metformin alone. AZD1656 is not approved by regulatory agencies and remains investigational. AstraZeneca has been the principal developer of AZD1656, and combination studies have assessed the safety, efficacy, and tolerability of AZD1656 as an add-on to metformin in this patient population[1][3][4][5].
