Overview

**AZD1656 + pioglitazone** is a combination therapy under investigation for the treatment of type 2 diabetes mellitus. AZD1656 is a glucokinase activator developed to enhance glucose-dependent insulin release by increasing the activity of glucokinase, a key enzyme in the regulation of carbohydrate metabolism. Pioglitazone is a thiazolidinedione that acts as an agonist of the peroxisome proliferator-activated receptor gamma (PPARγ), thereby improving insulin sensitivity. The combination is designed to leverage both improved insulin release (from AZD1656) and increased insulin sensitivity (from pioglitazone) to achieve better glycemic control in diabetes patients. A phase 1 clinical trial (cross-over design) evaluated the pharmacokinetics and safety of the combination in type 2 diabetic patients treated with metformin[1][7].
