AZD2811 + venetoclax is a combination therapy of two investigational anti-cancer agents. **AZD2811** is a nanoparticle-encapsulated, selective inhibitor of Aurora kinase B, an enzyme critical for mitosis, leading to impaired cell division and apoptosis. **Venetoclax** is a highly selective small molecule inhibitor of the anti-apoptotic protein BCL2, promoting programmed cell death in susceptible cancer cells. The combination is designed to synergistically induce apoptosis in cancer cells, particularly those resistant to single-agent therapy, by simultaneously targeting cell cycle regulatory pathways (Aurora kinase B) and apoptosis resistance mechanisms (BCL2). This combination shows promising preclinical activity in small cell lung cancer (SCLC) models with high BCL2 expression and acute myeloid leukemia (AML), with enhanced tumor regression and apoptosis compared to monotherapies. AZD2811 is developed by AstraZeneca; venetoclax was developed by AbbVie in partnership with Genentech/Roche.
