AZD4017 + prednisolone

AZD4017 + prednisolone is a combination therapy consisting of AZD4017, a selective 11β-hydroxysteroid dehydrogenase type 1 (11β-HSD1) inhibitor, and prednisolone, a glucocorticoid with anti-inflammatory properties. This combination aims to reduce the adverse metabolic effects of prednisolone while maintaining its anti-inflammatory benefits. AZD4017 works by inhibiting the conversion of inactive cortisone to active cortisol in tissues, thereby limiting prednisolone's adverse effects on metabolism, bone turnover, and blood pressure. Clinical trials have shown that co-administration of AZD4017 with prednisolone in men demonstrated **protective effects on markers of lipid metabolism and bone turnover**. The combination also **prevented prednisolone-induced suppression of bone formation biomarkers, reduction of insulin sensitivity, elevation of triglycerides, and night-time diastolic hypertension**.
