AZD4547 + docetaxel is a combination therapy investigated for the treatment of recurrent FGFR1-amplified squamous non-small cell lung cancer (NSCLC). AZD4547 is an orally bioavailable, potent, and selective small molecule inhibitor of the fibroblast growth factor receptors 1, 2, and 3 (FGFR1/2/3). Docetaxel is a taxane-based chemotherapy agent that promotes the assembly of tubulin into stable microtubules and inhibits their disassembly, leading to cell cycle arrest in the G2/M phase and subsequent apoptosis. This combination was evaluated in a Phase I/II clinical trial (NCT01795716) sponsored by the ECOG-ACRIN Cancer Research Group to determine if the dual inhibition of FGFR signaling and mitotic progression could improve outcomes in patients with FGFR-driven squamous NSCLC. Clinical results for this combination in FGFR1-amplified squamous NSCLC have generally shown modest efficacy.
