Overview
AZD5305 + enzalutamide is an investigational combination therapy for metastatic prostate cancer consisting of saruparib (AZD5305), a next-generation, highly selective PARP1 inhibitor, and enzalutamide, an androgen receptor inhibitor. AZD5305 inhibits PARP1-mediated DNA repair in cancer cells, thereby enhancing DNA damage and promoting cancer cell death, while enzalutamide blocks androgen receptor signaling required for prostate cancer growth. The combination targets both DNA repair and hormonal pathways and is primarily under clinical investigation for metastatic castration-resistant and castration-sensitive prostate cancer, with the aim of improving progression-free survival and tolerability compared to standard therapies[2][3][5][6].
