AZD5396 + AZD8076 is an investigational **DNA-encoded monoclonal antibody** cocktail for pre-exposure prophylaxis against COVID-19. It consists of synthetic plasmid DNA constructs encoding the heavy and light chains of two Fc-engineered anti-SARS-CoV-2 neutralizing antibodies, AZD5396 and AZD8076, which are modified DNA-delivered versions derived from the tixagevimab and cilgavimab antibody pair and ultimately from parental clones COV2-2196 and COV2-2130. After **intramuscular injection** followed by **CELLECTRA electroporation**, host muscle cells produce the antibodies in vivo, generating sustained systemic exposure and spike receptor-binding-domain neutralizing activity. The program has been developed by INOVIO, with the encoded antibodies originating from AstraZeneca's anti-SARS-CoV-2 antibody work, and has been evaluated clinically in a Phase 1 study in healthy adults.
