AZD8205 + AZD9574 + AZD2936 is a triplet combination therapy being developed by AstraZeneca for the treatment of advanced or metastatic solid tumors, including breast, ovarian, endometrial, and biliary tract cancers. The regimen consists of three distinct agents: AZD8205 (puxitatug samrotecan), an antibody-drug conjugate (ADC) targeting B7-H4 with a topoisomerase I inhibitor payload; AZD9574, a brain-penetrant, selective PARP1 inhibitor that induces synthetic lethality in DNA repair-deficient cells; and AZD2936 (rilvegostomig), a bispecific antibody targeting the immune checkpoints PD-1 and TIGIT. This combination is designed to synergistically target tumor cells through direct cytotoxicity, inhibition of DNA damage response, and reactivation of the anti-tumor immune response. It is currently being evaluated in Sub-Study 4 of the Phase I/IIa BLUESTAR master protocol trial (NCT05123482).
