Overview

AZD8421 + camizestrant + abemaciclib is an investigational oral combination regimen developed for the treatment of advanced or metastatic hormone receptor-positive, HER2-negative breast cancer and potentially other solid tumors. AZD8421 is a selective cyclin-dependent kinase 2 (CDK2) inhibitor. Camizestrant (AZD9833) is a next-generation oral selective estrogen receptor degrader (SERD), and abemaciclib is an oral inhibitor of cyclin-dependent kinases 4 and 6 (CDK4/6). This triple combination targets cell cycle progression and estrogen receptor signaling, aiming to overcome resistance mechanisms in patients previously treated with CDK4/6 inhibitors and endocrine therapies. Early clinical evaluation is ongoing to assess safety, tolerability, and antitumor activity.
