Overview

**AZD8421 + camizestrant + palbociclib** is an experimental oral combination therapy being studied primarily for estrogen receptor-positive (ER+), human epidermal growth factor receptor 2-negative (HER2-) advanced or metastatic breast cancer. AZD8421 is a highly selective cyclin-dependent kinase 2 (CDK2) inhibitor, camizestrant is a next-generation oral selective estrogen receptor degrader (SERD), and palbociclib is a CDK4/6 inhibitor. The rationale for this combination is to overcome or delay resistance to current standard-of-care therapies targeting ER and cell cycle pathways. The combination is under clinical investigation in the CYCAD-1 trial, which assesses its safety, tolerability, pharmacokinetics, and preliminary efficacy in patients who have progressed after prior CDK4/6 inhibitor therapy[1][4][7][9].
