AZD9574 + datopotamab deruxtecan is an experimental combination therapy under investigation for advanced solid tumors. AZD9574 is a selective, blood-brain barrier–penetrant PARP1 (poly ADP-ribose polymerase 1) inhibitor, designed to exploit homologous recombination repair deficiency in cancer cells and shows strong preclinical efficacy, particularly in brain tumors. Datopotamab deruxtecan (Dato-DXd, brand name Datroway) is an antibody-drug conjugate (ADC) consisting of a Trop-2–targeting monoclonal antibody (datopotamab) linked to a topoisomerase I inhibitor (DXd payload), recently FDA-approved for unresectable or metastatic, HR-positive, HER2-negative breast cancer, and accelerated approval for EGFR-mutated non-small cell lung cancer. The combination aims to harness the DNA-damage effects of both agents, potentially increasing antitumor activity in a variety of advanced solid tumors.
