Overview

This combination drug is an investigational regimen being studied primarily in metastatic colorectal cancer. It consists of AZD9592, a novel antibody-drug conjugate targeting EGFR and c-MET, combined with 5-fluorouracil (a thymidylate synthase inhibitor antimetabolite), bevacizumab (a VEGF-A-targeting monoclonal antibody), and leucovorin (a folinic acid that enhances 5-FU activity). AZD9592 is under early-phase clinical evaluation to assess safety, tolerability, and anti-tumor activity in advanced solid tumors, including colorectal cancer, when used together with established chemotherapy and targeted agents. The constituent drugs have individually shown efficacy in solid malignancies, particularly gastrointestinal cancers, with bevacizumab + fluorouracil + leucovorin forming a backbone regimen for metastatic colorectal cancer[5][7][2][3].
