Overview

TAK-536TCH is a fixed-dose triple combination therapy developed by Takeda for the management of hypertension. It combines three antihypertensive agents with complementary mechanisms of action: azilsartan medoxomil (an angiotensin II receptor blocker), amlodipine besylate (a dihydropyridine calcium channel blocker), and hydrochlorothiazide (a thiazide diuretic). Azilsartan inhibits the renin-angiotensin-aldosterone system by selectively blocking the AT1 receptor, amlodipine promotes vasodilation by inhibiting calcium influx into vascular smooth muscle, and hydrochlorothiazide reduces blood volume by inhibiting sodium reabsorption in the distal renal tubules. This triple combination is designed to provide enhanced blood pressure control in patients who do not achieve target levels with dual therapy. Clinical development has included Phase 1 food effect studies and Phase 3 efficacy trials in Japanese populations.
