Overview

A combination of **azithromycin**, a macrolide antibiotic that inhibits bacterial protein synthesis by binding to the 50S ribosomal subunit, and **colchicine**, a microtubule inhibitor used primarily in the management of gout and familial Mediterranean fever. This combination is not marketed as a fixed co-formulated product, but both drugs may be administered concurrently in clinical settings. Colchicine acts by inhibiting microtubule polymerization, leading to disruption of neutrophil activity and reductions in inflammation. Azithromycin is primarily bacteriostatic, though it may exhibit bactericidal activity at high concentrations or against certain organisms. These drugs have overlapping metabolic pathways involving P-glycoprotein; however, available studies indicate that, unlike clarithromycin, azithromycin does not significantly increase colchicine exposure, and dose adjustment is generally not required when used together in patients with normal renal and hepatic function.
