Overview

This combination therapy consists of the macrolide antibiotic azithromycin and the synthetic corticosteroid dexamethasone. It is being evaluated by Assiut University in a Phase 4 clinical trial for the prevention of postoperative nausea and vomiting (PONV) in women undergoing elective Caesarean section under spinal anesthesia. Azithromycin, beyond its antimicrobial properties, acts as a motilin receptor agonist, which stimulates gastrointestinal motility (prokinetic effect) and may help mitigate nausea. Dexamethasone is a well-established antiemetic agent that likely functions by inhibiting prostaglandin synthesis or modulating the solitary tract nucleus in the central nervous system. The combination aims to leverage these distinct mechanisms to provide superior PONV prophylaxis compared to standard monotherapy.
