Overview

Azithromycin + pyrimethamine is a combination of two antimicrobial agents — azithromycin, a macrolide antibiotic, and pyrimethamine, an antiparasitic agent. This combination has been studied for the treatment of acute toxoplasmosis, particularly in immunocompromised patients, where monotherapy may be less effective. In animal studies and small clinical trials, this combination has demonstrated a *remarkable synergistic effect*, markedly reducing parasite burdens, decreasing relapses, and lowering mortality when compared to either drug alone. Azithromycin inhibits protein synthesis by binding to the 50S subunit of the bacterial ribosome, while pyrimethamine inhibits dihydrofolate reductase, disrupting folate synthesis critical for protozoal DNA replication[1][3][5][7].
