Azvudine + rilpivirine is a combination of two small molecule antiretroviral drugs used primarily in the treatment of HIV-1 infection. **Azvudine** is a nucleoside reverse transcriptase inhibitor (NRTI) and also acts by restoring cytidine deaminase expression, offering broad-spectrum antiviral activity against HIV, HCV, HBV, and certain other viruses. **Rilpivirine** is a non-nucleoside reverse transcriptase inhibitor (NNRTI) that non-competitively binds to the reverse transcriptase enzyme, blocking RNA and DNA-dependent DNA polymerase activities crucial for HIV-1 replication. Both drugs are designed to prevent viral replication via inhibition of reverse transcriptase, but each targets different aspects of the enzyme’s activity, providing complementary effects. The combination is primarily developed and used for the management of HIV-1, particularly for patients with high viral loads, and is considered synergistic when used with other antiretroviral agents. Azvudine was developed by Henan Sincere Biotech and has received conditional regulatory approval for HIV-1 and COVID-19 treatment in China. Rilpivirine is developed and manufactured by Janssen Pharmaceuticals and is widely approved for HIV-1 treatment in combination regimens[1][3][4][5][9].
