Overview

MC10029 (BAFFR CAR-T) is an autologous chimeric antigen receptor (CAR) T-cell therapy targeting the B-cell activating factor receptor (BAFF-R), a member of the tumor necrosis factor receptor superfamily essential for B-cell survival and maturation. Developed by the Mayo Clinic, the therapy involves engineering a patient's own T cells using a lentiviral vector to express a CAR construct containing an scFv derived from a novel anti-BAFF-R monoclonal antibody. BAFF-R is consistently expressed on malignant B cells, including those that have lost CD19 expression, making MC10029 a potential treatment for patients with relapsed or refractory B-cell malignancies who have failed CD19-targeted therapies. It is currently being evaluated in Phase 1 clinical trials (NCT06191887) for conditions such as chronic lymphocytic leukemia (CLL) and other B-cell hematologic malignancies.
