These agents utilize cyclic peptides as high-affinity targeting ligands for B7-H3 (CD276) and DLL3 (Delta-like protein 3), respectively. B7-H3 is an immune checkpoint molecule widely expressed in solid tumors, while DLL3 is a Notch ligand frequently found in neuroendocrine cancers like small cell lung cancer. By conjugating these peptides to radioisotopes, the compounds deliver targeted radiation to malignant cells. The discovery process, led by researchers at Full-Life Technologies, employed phage display libraries and AlphaFold3 structural modeling to optimize binding kinetics and selectivity, aiming to overcome the limitations of traditional ligand discovery in the RDC space.
