Overview

BA3021 + PD-1 inhibitor is a **combination therapy** consisting of BA3021 (ozuriftamab vedotin), a conditionally active biologic (CAB) antibody-drug conjugate targeting receptor tyrosine kinase orphan receptor 2 (ROR2), and a programmed death-1 (PD-1) inhibitor. BA3021 is a humanized IgG1 monoclonal antibody conjugated to the cytotoxic payload monomethyl auristatin E (MMAE) via a cleavable linker. The CAB design reduces off-tumor toxicity by selectively binding ROR2 under acidic tumor microenvironment conditions. This combination aims to enhance antitumor immune responses and is under development for advanced solid tumors, mainly recurrent or metastatic squamous cell carcinoma of the head and neck, non-small cell lung cancer, melanoma, triple-negative breast cancer, and soft tissue sarcoma[1][3][4][5].
