**BA3071 + pembrolizumab** is an investigational combination therapy for advanced solid tumors, comprising BA3071—a conditionally active biologic (CAB) anti–CTLA-4 monoclonal antibody—and pembrolizumab, an approved monoclonal antibody checkpoint inhibitor targeting programmed cell death protein 1 (PD-1). BA3071 blocks CTLA-4 interaction with its ligands (CD80 and CD86) and is designed for activation within the acidic tumor microenvironment to minimize off-tumor toxicity. Pembrolizumab inhibits PD-1 to enhance antitumor immune responses by preventing T-cell inhibition via PD-L1. The combination aims to augment immune-mediated tumor destruction with improved tolerability over traditional CTLA-4/PD-1 blockade. BA3071 is developed for use in solid tumors, including melanoma, carcinoma, and urothelial carcinoma.
