BA3071 + pembrolizumab + pemetrexed is an investigational combination therapy that merges **BA3071** (a novel immunotherapy agent, presumed to be an anti–CTLA-4 antibody based on its development pipeline), **pembrolizumab** (a programmed cell death protein 1 [PD-1] inhibitor immunotherapy), and **pemetrexed** (a chemotherapy agent and antifolate antineoplastic). - **BA3071** is being developed as a next-generation, tumor microenvironment–activated anti–CTLA-4 antibody designed to reduce systemic toxicity associated with current anti–CTLA-4 therapies (such as ipilimumab), and to enhance the anti-tumor immune response in synergy with anti–PD-1 agents. - **Pembrolizumab** is a monoclonal antibody that blocks PD-1, releasing T cells from checkpoint-mediated immunosuppression to restore anti-tumor immunity. - **Pemetrexed** is an anti-cancer chemotherapy primarily used for non-squamous non–small-cell lung cancer (NSCLC), which inhibits multiple folate-dependent enzymes involved in purine and pyrimidine synthesis, curbing DNA and RNA reproduction in rapidly-dividing cells. The combination approach is under investigation for synergistic immunologic activation, enhancing both direct cytotoxic and immune-mediated tumor cell killing, with a primary focus on advanced/metastatic non-squamous NSCLC lacking actionable driver mutations.
