This is an investigational combination immunotherapy regimen consisting of Bacillus Calmette-Guerin (BCG) and interferon alpha-2b, specifically evaluated by the National University Hospital, Singapore, for the treatment of high-risk superficial (non-muscle invasive) bladder cancer. BCG is a live attenuated strain of Mycobacterium bovis that, when administered intravesically, induces a robust local inflammatory and immune response characterized by the activation of macrophages, T-cells, and the production of various cytokines. Interferon alpha-2b is a recombinant cytokine that further modulates the immune system, enhancing the anti-tumor activity and potentially allowing for a reduction in the BCG dose to mitigate toxicity. Clinical trials conducted by the National University Hospital have demonstrated that this combination therapy provides superior recurrence-free survival and significantly lower local and systemic toxicity compared to standard-dose BCG monotherapy in patients with aggressive superficial bladder cancer.
