Overview

Baclofen + tizanidine is a combination of two centrally acting muscle relaxants used for the management of spasticity, particularly in conditions such as multiple sclerosis or spinal cord injury. Baclofen acts primarily as a gamma-aminobutyric acid (GABA) B receptor agonist, inhibiting excitatory neurotransmitter release and reducing muscle spasticity. Tizanidine is an alpha-2 adrenergic receptor agonist that inhibits presynaptic motor neurons, decreasing muscle tone and frequency of spasms. The combination may be used to achieve better control of spasticity at lower doses of each agent, potentially minimizing side effects associated with higher doses of either drug alone. However, additive sedative effects are possible when these drugs are combined[1][4][7].
