Drug intelligence / Profile preview
Bacteriophage + cerium oxide (specifically studied as P510@CeO2) is an engineered biohybrid nanomedicine developed for the treatment of carbapenem-resistant *Klebsiella pneumoniae* (CRKP)-associated inflammatory bowel disease (IBD). It consists of a lytic bacteriophage (such as P510) targeting the ST11 KL64 strain of CRKP, covalently conjugated to catalytic cerium oxide ($CeO_2$) nanoparticles. The bacteriophage component provides precise targeting of CRKP by recognizing bacterial capsular polysaccharides and outer membrane proteins, leading to bacterial lysis. The cerium oxide nanoparticle component acts as a catalytic reactive oxygen species (ROS) scavenger and anti-inflammatory agent, alleviating oxidative stress and inflammation in the inflamed colon. This dual-action approach also downregulates capsule-encoding genes and inhibits efflux pumps to prevent the emergence of phage-resistant mutants, restores intestinal microbiota balance, and repairs the intestinal barrier.
