Baicalein + oxaliplatin is a combination therapeutic delivered via a nanoparticle system (B/O@NPs) designed to treat pancreatic ductal adenocarcinoma (PDAC). The system utilizes a dual-mechanism approach to induce pyroptosis, a form of programmed cell death, to remodel the immunosuppressive tumor microenvironment. Baicalein acts by inhibiting NF-κB nuclear translocation, leading to mitochondrial dysfunction and a reactive oxygen species (ROS) burst that activates the NLRP3/caspase-1/GSDMD pathway. Simultaneously, oxaliplatin, a platinum-based chemotherapy agent, triggers caspase-3-mediated cleavage of GSDME. This synergistic activation of dual gasdermin pathways promotes immunogenic cell death (ICD), increasing the infiltration of mature dendritic cells and CD8+ T cells while improving the M1/M2 macrophage ratio. Preclinical studies in PDAC models have demonstrated that this nanoplatform significantly inhibits tumor growth and mitigates systemic toxicity compared to free oxaliplatin.
