BAL0891 + carboplatin is an experimental combination therapy being investigated for cancer treatment, particularly advanced solid tumors and relapsed or refractory acute myeloid leukemia. BAL0891 is a first-in-class, dual inhibitor of threonine tyrosine kinase (TTK, also known as Mps-1) and polo-like kinase 1 (PLK1), key kinases that regulate the spindle assembly checkpoint (SAC) in mitosis. The combination with carboplatin, a platinum-based DNA-damaging chemotherapy, is being evaluated for synergistic anti-tumor effects. BAL0891 demonstrates prolonged inhibition of TTK and transient inhibition of PLK1, disrupting the SAC, inducing aberrant mitotic exit, and leading to tumor cell death. In preclinical models, synergy and improved anti-tumor results have been seen when carboplatin is given prior to BAL0891. This combination is currently under investigation in phase 1 clinical trials for indications such as triple-negative breast cancer, ovarian cancer, and other advanced solid tumors[1][2][3].
