BAL0891 + paclitaxel is a combination therapy under investigation for advanced and metastatic solid tumors, notably including triple-negative breast cancer (TNBC). BAL0891 is a novel, small molecule kinase inhibitor that targets both TTK (also known as MPS1) and PLK1, disrupting mitotic checkpoint (spindle assembly checkpoint, SAC) signaling and leading to aberrant mitotic progression and tumor cell death. Paclitaxel is a standard-of-care (SoC) chemotherapeutic agent that stabilizes microtubules, impairing mitosis. Preclinical and early clinical data show that this combination exhibits strong synergy and tumor regression, outperforming monotherapies. The combination is currently being evaluated in phase 1 dose-escalation and expansion studies for efficacy and safety in solid tumors, including TNBC and gastric cancer[1][2][3].
