balovaptan + vasopressin is a combination of the selective small molecule vasopressin V1A receptor antagonist balovaptan (RG7314) and the neuropeptide arginine vasopressin (AVP). This combination was primarily utilized in clinical research settings, specifically in a Phase 1 proof-of-mechanism study by Hoffmann-La Roche to investigate the effects of RG7314 on brain activity and connectivity in response to an intranasal vasopressin challenge. The underlying hypothesis was that modulating the vasopressin pathway could improve social communication and interaction in individuals with autism spectrum disorder (ASD). While balovaptan received Breakthrough Therapy Designation from the FDA in 2018, subsequent clinical trials failed to demonstrate statistically significant improvements in socialization and communication in children and adolescents, leading to the cessation of its development for this indication.
