**BAT8010 + BAT1006** is an investigational combination therapy for advanced and metastatic solid tumors, particularly in HER2-positive cancers. BAT8010 is a HER2-targeting antibody-drug conjugate (ADC) that uses a novel cleavable linker to conjugate an exatecan payload (a topoisomerase I inhibitor) with a high drug-to-antibody ratio. BAT1006 is a humanized, afucosylated monoclonal antibody targeting a non-overlapping HER2 epitope, designed for enhanced antibody-dependent cellular cytotoxicity (ADCC). BAT1006 blocks HER2 heterodimerization with EGFR, HER3, and HER4, inhibiting downstream signaling. Used together, the drugs aim to improve therapeutic efficacy and overcome resistance in HER2-positive cancers, including those previously treated with HER2-ADCs. Early clinical data suggest a favorable safety profile (mainly manageable hematological toxicity) and promising antitumor activity, especially in metastatic breast and gastric cancer, including patients with HER2-low expression or resistance to previous HER2-targeted therapies[1][3][4][5][7].
