Bavituximab + durvalumab is an investigational combination therapy composed of two monoclonal antibodies with distinct immunotherapeutic mechanisms. Bavituximab is a chimeric IgG1 monoclonal antibody that targets phosphatidylserine (PS), an immunosuppressive phospholipid exposed on the surface of tumor cells and tumor vasculature under stress conditions. By binding to PS, bavituximab reverses the immunosuppressive environment in tumors and increases T-cell activation within the tumor microenvironment. Durvalumab is a human monoclonal antibody directed against programmed cell death ligand 1 (PD-L1). It blocks PD-L1 from interacting with its receptors PD-1 and CD80 on T-cells, thereby preventing immune evasion by tumors and enhancing anti-tumor immune responses. The combination aims to synergistically enhance anti-tumor immunity by both reversing local immunosuppression (bavituximab) and blocking checkpoint inhibition (durvalumab). This regimen has been evaluated in early-phase clinical trials for multiple solid tumors[1][2][3].
