Bavituximab + paclitaxel is a combination therapy consisting of bavituximab, a chimeric monoclonal antibody that targets phosphatidylserine (PS) on tumor vasculature, and paclitaxel, a microtubule-stabilizing chemotherapeutic agent. Bavituximab binds to PS exposed on the surface of tumor blood vessels under stress conditions such as hypoxia, blocking immunosuppressive signals and promoting antitumor immunity through effects on monocytes and myeloid-derived suppressor cells. It also induces antibody-dependent cellular cytotoxicity against tumor-associated endothelial cells. Paclitaxel disrupts cell division by stabilizing microtubules, leading to apoptosis in rapidly dividing cancer cells. The combination has been investigated in multiple phase I/II clinical trials for advanced solid tumors including metastatic breast cancer, non-small cell lung cancer (NSCLC), and gastric/gastroesophageal junction (GEJ) cancers[1][2][8]. The regimen has shown promising response rates and manageable toxicity profiles.
