**Bavituximab + taxane** is a combination therapy under investigation primarily for advanced non-small-cell lung cancer (NSCLC). Bavituximab is an unconjugated, chimeric IgG1 monoclonal antibody targeting phosphatidylserine (PS)—an immunosuppressive phospholipid exposed on the outer leaflet of the plasma membrane in the tumor microenvironment, especially under stress conditions like hypoxia. Bavituximab binds to PS in the presence of β2-glycoprotein 1 (β2GP1), promoting tumor vessel destruction and reversal of immunosuppression, repolarizing myeloid-derived suppressor cells and M2 macrophages to boost pro-inflammatory and cytotoxic anti-tumor immune responses. The taxane component (such as docetaxel or paclitaxel) is a cytotoxic chemotherapy agent that stabilizes microtubules and inhibits cell division. Clinical trials have evaluated this combination as first-line or second-line treatment in advanced NSCLC. The combination has shown manageable safety but has not demonstrated a significant improvement in overall survival compared to taxane alone based on phase III studies[1][3][4][6].
