Overview

BAY 3605349 + VVD-159642 is a combination of two investigational oral small molecule inhibitors, BAY 3605349 (also known as VVD-159642) targeting the RAS-PI3Kα signaling pathway and an associated compound from Vividion Therapeutics, a Bayer subsidiary. VVD-159642 selectively blocks oncogenic RAS activation of phosphatidylinositol 3-kinase alpha (PI3Kα), a key pathway in solid tumor development, while avoiding disruption of normal cellular RAS-PI3Kα function, potentially offering tolerability advantages over prior attempts. Developed using Vividion's covalent chemoproteomics platform, it is being evaluated in Phase I trials as monotherapy and in combinations with KRAS G12C inhibitor sotorasib or MEK inhibitor trametinib for advanced solid tumors with RAS mutations, EGFR/HER2 overexpression, or related alterations, including pancreatic, colorectal, and lung cancers.[1][3][4]
