Overview

BAY2965501 + pembrolizumab is an investigational combination therapy being studied for the treatment of advanced solid tumors. BAY2965501 is a highly selective and potent oral inhibitor of diacylglycerol kinase zeta (DGKζ), developed to enhance anti-tumor immune responses by activating T cells and natural killer (NK) cells. It works by overcoming immunosuppressive signals in the tumor microenvironment that lead to T cell exhaustion. Pembrolizumab is a well-established monoclonal antibody targeting programmed death receptor 1 (PD-1), functioning as an immune checkpoint inhibitor to restore T cell activity against cancer cells. The combination aims to synergistically boost anti-tumor immunity by both relieving inhibitory signaling on T cells (via DGKζ inhibition) and blocking PD-1-mediated suppression. This regimen is currently under investigation in phase 1 clinical trials for patients with advanced or metastatic solid tumors who have limited treatment options[3][6][9].
