This combination therapy consists of bazedoxifene, a third-generation selective estrogen receptor modulator (SERM), and eldecalcitol, an active vitamin D3 analog. Bazedoxifene (Viviant) acts as an estrogen receptor agonist in bone, effectively inhibiting bone resorption and maintaining bone mineral density without stimulating the endometrium or breast tissue. Eldecalcitol (Edirol) is a vitamin D receptor agonist that enhances intestinal calcium absorption and further suppresses bone resorption. The combination is specifically being investigated by Shinshu University in a Phase 2 clinical trial as a sequential therapy for Japanese patients with osteoporosis who have previously received denosumab, aiming to prevent the rapid bone loss and increased fracture risk often observed after denosumab discontinuation.
