Overview

BB-031 is a first-in-class, high-affinity RNA aptamer that targets the A1 domain of von Willebrand Factor (vWF). Developed by Basking Biosciences, it is designed for the acute treatment of ischemic stroke by inhibiting vWF-mediated platelet adhesion and arterial thrombosis. By specifically blocking the interaction between vWF and the platelet receptor GPIba, BB-031 prevents thrombus formation and promotes recanalization. A unique feature of this therapeutic system is BB-025, a specific reversal agent consisting of a complementary oligonucleotide. BB-025 is designed to rapidly bind and neutralize BB-031 through Watson-Crick base pairing, providing a "safety switch" to restore normal vWF function and manage potential bleeding complications. This reversible antithrombotic approach aims to provide a safer alternative to traditional non-reversible thrombolytics and expand the treatment window for stroke patients.
