BCG022 is a novel bispecific antibody-drug conjugate (bsADC) being developed by Biocytogen Pharmaceuticals. It consists of a fully human bispecific antibody, generated using the proprietary RenLite® mouse platform, that simultaneously targets HER3 (receptor tyrosine-protein kinase erbB-3) and MET (hepatocyte growth factor receptor). The antibody is conjugated to the cytotoxic payload monomethyl auristatin E (MMAE), a microtubule inhibitor, although other payloads such as BLD1102 have also been evaluated. BCG022 is designed to treat multiple solid tumor types where HER3 and MET are co-expressed, including gastric cancer, colorectal cancer, breast cancer, non-small-cell lung cancer (NSCLC), hepatocellular carcinoma, and pancreatic cancer. By targeting both HER3 and MET, the drug aims to overcome therapeutic resistance, as these receptors often serve as bypass resistance mechanisms to EGFR-targeted therapies. BCG022 is currently in preclinical development and has demonstrated anti-tumor efficacy in various cell-derived and patient-derived xenograft models.
