**BCMA CAR-T cells** are a type of autologous or allogeneic cellular immunotherapy where patient (or donor) T cells are genetically engineered to express a chimeric antigen receptor (CAR) that specifically recognizes B-cell maturation antigen (BCMA), a transmembrane protein highly expressed on malignant plasma cells in multiple myeloma. Upon reinfusion, these CAR-T cells bind to BCMA-expressing cells, activating the T cells to exert cytotoxic effects and kill the cancer cells. BCMA-directed CAR-T cell therapy has demonstrated efficacy in relapsed and refractory multiple myeloma, with approved products in the U.S. including idecabtagene vicleucel and ciltacabtagene autoleucel. The main mechanism of action is the direct recognition and destruction of BCMA-positive malignant plasma cells, with secondary effects including cytokine release and immune activation. This therapy is typically administered via a single intravenous infusion after lymphodepleting chemotherapy[3][4][5][8][9][10].
