This dual-targeting chimeric antigen receptor (CAR) T-cell therapy is a biologic immunotherapy designed to simultaneously target B-cell maturation antigen (BCMA) and G protein-coupled receptor class C group 5 member D (GPRC5D). Developed by Beijing GoBroad Hospital, the therapy involves the genetic modification of a patient's T cells to express receptors against both antigens, a strategy intended to mitigate antigen escape-mediated relapse, which is a frequent challenge in single-target BCMA CAR-T treatments. It is specifically being investigated for the treatment of relapsed or refractory multiple myeloma (RRMM), with a particular focus on patients presenting with extramedullary infiltration (EMI), a high-risk manifestation associated with poor prognosis. The therapy is currently undergoing Phase 1 clinical evaluation to assess its safety, tolerability, and preliminary efficacy in heavily pretreated patient populations.
