BCMA ScFv-CD3 ScFv-Fc (tetravalent) is a bispecific T-cell engager (BiTE) construct developed by ProMab Biotechnologies for the treatment of multiple myeloma. This specific molecular architecture utilizes a tetravalent format, incorporating humanized single-chain variable fragments (scFv) targeting B-cell maturation antigen (BCMA) and CD3, fused to an Fc region. The humanized BCMA component is derived from clone 4C8A. By simultaneously binding to BCMA on malignant plasma cells and CD3 on T cells, the antibody facilitates the formation of an immunological synapse, leading to T-cell activation and redirected cytotoxic lysis of the tumor cells. Preclinical studies presented at AACR 2022 demonstrated that this tetravalent design, along with other bispecific formats, exhibits high affinity for BCMA-positive cells and potent dose-dependent cytotoxicity in multiple myeloma cell lines (such as MM1S and H929) and xenograft mouse models.
