Drug intelligence / Profile preview
BCMA-targeting bispecific T-cell engagers (TCEs) are a class of immunotherapeutic agents designed to treat multiple myeloma and autoimmune diseases by redirecting the patient's own T cells to eliminate malignant or autoreactive plasma cells. These bispecific molecules simultaneously bind to B-cell maturation antigen (BCMA), which is highly and selectively expressed on the surface of multiple myeloma cells and plasma cells, and CD3, a component of the T-cell receptor complex. This dual binding facilitates the formation of an immunological synapse, leading to T-cell activation, proliferation, and the release of cytotoxic perforins and granzymes that induce apoptosis in the BCMA-expressing cells. Several BCMA-targeting TCEs, such as teclistamab, elranatamab, and linvoseltamab, have been developed and approved or are in clinical development.
