BDC-3042 + cemiplimab is an investigational combination immunotherapy for advanced malignancies, currently under clinical development. BDC-3042 is a novel, agonist monoclonal antibody targeting dectin-2, a C-type lectin receptor predominantly expressed on tumor-associated macrophages (TAMs). Dectin-2 activation by BDC-3042 is intended to reprogram TAMs from an immunosuppressive to an immunostimulatory phenotype, promoting an enhanced anti-tumor immune response in the tumor microenvironment. Cemiplimab is a fully human monoclonal antibody PD-1 checkpoint inhibitor that blocks the programmed death-1 (PD-1) receptor on T cells, restoring and amplifying anti-tumor immune activity. The combination aims to leverage reprogramming of TAMs together with checkpoint inhibition to provide synergistic anti-tumor effects. The primary clinical indications under investigation include advanced solid tumors, such as non-small cell lung cancer, melanoma, triple-negative breast cancer, renal cell carcinoma, colon cancer, head and neck cancer, and ovarian cancer.
